Herein, the real-time and rapid detection of pesticide residues in agricultural products is of great significance for global food and environmental safety. Carbon quantum dots possess excellent fluorescence properties, adjustable emission wavelength, excellent biocompatibility and significant optical stability, which can be used to construct fluorescent probes for the detection of residual pesticides. This review briefly expounds the advantages of carbon quantum dots based fluorescent probe for pesticide detection, systematically analyzes the construction strategies of carbon quantum dots based fluorescent probe for residual pesticide detection via metal ions, nanoparticles, various enzymes and various antibodies, and its application in residual pesticide detection has been summarized and prospected. The conclusion of this paper will favor for researchers to reasonably design more excellent carbon quantum dots based fluorescent probes for residual pesticide detection. 碳量子点 农药检测 荧光探针 carbon quantum dots pesticide detection fluorescent probe 0　 引 言 1 农药（pesticide）是农业上用于防治病虫害及调节植物生长的化学物质，因此也是水果、蔬菜及其他农作物中最重要的残留毒物，是严重及长期存在的环境威胁 ［ 1 1 ， 2 2 ］ 。基于不同的化学组成，农药可以分为5类：有机氯（organochlorine）、有机磷（organophosphorus）、氨基甲酸盐（carbamate）、除虫菊素（pyrethrin）、拟除虫菊素（pyrethroid）类化合物。有机磷化合物在过去的几十年里被广泛并持续地用作杀虫剂、杀蠕虫剂、杀线虫剂、杀真菌剂和除草剂，因此全球使用的农药中占比最高的为有机磷类，大约为38% ［ 3 3 ， 4 4 ］ 。有机磷农药暴露造成的神经毒性主要表现在抑制乙酰胆碱酯酶（acetylcholinesterase，AChE）的活性，该典型水解酶对于中枢神经系统的正常工作非常重要 ［ 5 5 ］ 。几乎所有的有机磷农药都可以通过对AChE的磷酸化来抑制其活性，使AChE无法催化乙酰胆碱的水解，并引发乙酰胆碱的聚集，导致胆碱毒性 ［ 6 6 ］ 。其他类型的农药也会损伤人体免疫系统，导致人体激素紊乱以及耐药性等问题 ［ 7 7 ］ 。因此，考虑到农药的潜在毒性以及农药的残留富集性，构建面向残留农药的高灵敏度、高选择性的定量检测方法对于环境、食物的安全及人类的健康非常重要。 许多传统的方法，如气相色谱（GC） ［ 8 8 ］ 、高效液相色谱（HPLC） ［ 9 9 ］ 及质谱（MS） ［ 10 10 ］ 等均在农药的定量检测方面有广泛的应用。但这些方法复杂的操作步骤及高耗时等问题严重影响了其对农药的实时监测。荧光传感技术具有简便、快速、低成本以及高通量等特性，在监测残留农药领域的应用吸引了广泛的关注 ［ 11 11 ］ 。荧光检测收集的光学信号直接来源于发光材料，所以寻找荧光量子产率高及近红外发射的发光材料对于构建面向有机磷农药的高灵敏诊断平台非常重要。有机荧光染料、荧光蛋白、稀土纳米粒子以及半导体量子点具有许多不可避免的缺点，如光漂白、价格昂贵、稳定性较低以及毒性高等，限制了其实际应用 ［ 12 12 ］ 。与其他发光材料比较而言，碳量子点（carbon quantum dots，CDs）具有独特的化学结构及光学性质，如优异的荧光性能、可调的发射波长、良好的生物相容性及显著的光稳定性。同时，CDs表面含有多种官能团，如氨基、羧基、羟基及羰基等，为其功能化应用提供了基本条件。基于以上优点，CDs在生物传感、肿瘤治疗，尤其是环境污染监测 ［ 13 13 ， 14 14 ］ 领域得到越来越广泛的应用。 本文首先简单介绍了用于残留农药检测的CDs荧光探针的传感机制，然后从基于金属离子、纳米颗粒、天然酶及抗体4个角度，结合具体实例分析了用于残留农药检测的CDs荧光探针构建策略，并分别讨论了每种策略的机制及优缺点，最后对用于残留农药检测的CDs荧光检测技术进行了总结及展望。 1　 CDs荧光探针的传感机制 1 CDs具有良好的水溶性、低毒性及表面易修饰等优点，被广泛应用于构建高灵敏度的、检测残留农药的荧光探针。用于残留农药检测的CDs荧光探针传感机制可以分为3类： 1） CDs表面的活性基团（如合成过程中形成的羟基、羧基、氨基等）与残留农药直接作用； 2） 通过荧光“淬灭-恢复”（“Off-On”）的机制，将CDs与发光团、底物或淬灭剂结合后，再与残留农药作用； 3） 将CDs与某些特异性配体（如抗体、适配体、多肽及氨基酸）偶联，再与残留农药发生特异性结合。 根据残留农药浓度与CDs荧光信号改变之间的对应关系，可以构建用于目标农药检测的高效及灵敏的荧光探针。另外，将CDs优异的荧光性质与常用的多种传感机制（如光诱导的电子转移、荧光共振能量转移（FRET）、分子内电荷转移（ICT）、聚集诱导发光（AIE）及内过滤效应（IFE））结合，或通过功能化策略，与其他传感材料（酶及淬灭剂）结合，还可以监测荧光信号的淬灭、增敏及比率型响应行为。这些技术的运用，使用于残留农药检测的CDs荧光探针在灵敏度及选择性方面得到巨大和快速的发展。 2　 CDs荧光探针 1 根据改变CDs荧光信号的物质的不同，用于残留农药检测的CDs荧光探针可以分为以下4类：基于金属离子、纳米颗粒、天然酶及抗体的CDs荧光探针。 2.1　基于金属离子的CDs荧光探针 2 一些金属离子可以有效地淬灭CDs的荧光 ［ 15 15 ］ 。当农药与金属离子之间的亲和力大于金属离子与CDs之间的亲和力时，就可以利用这些金属离子作为CDs信号的淬灭剂，基于金属离子、CDs及目标农药之间的相互作用，构建“Off-On”型的荧光探针。 基于这一原则，Wang等 ［ 16 16 ］ 以硫脲作为碳源、二甘醇作为溶剂合成CDs，再以Cu 2+ 作为CDs的淬灭剂，利用Cu 2+ 与CDs表面基团及草甘膦之间的竞争性结合，实现了对草甘膦的增敏型检测。在最佳实验条件下，该方法的线性检测范围为0.03~10 μg/mL，检出限为16 ng/mL ［ 16 16 ］ 。近年来，可用于同时检测多种物质的多重传感平台吸引了科学界的广泛关注。Chen等 ［ 17 17 ］ 对柠檬酸及丙烯酰胺的甲酰胺溶液进行溶剂热处理，成功获取了发射红色荧光的CDs。CDs的荧光可以选择性地被Hg 2+ 淬灭，而农药福美锌可以与Hg 2+ 形成更稳定的复合物，使CDs的荧光恢复，据此建立了福美锌的检测方法。在该方法中， 红色荧光CDs的使用可以消除复杂体系中自发荧光的背景干扰，增强检测的灵敏度，福美锌的检出限为0.55 μg/mL。Wang等 ［ 18 18 ］ 则使用辛硫酸合成了一种硫掺杂的CDs（SCDs），并利用SCDs构建了能同时检测Hg 2+ 及甲基硫菌灵（thiophanate methyl，TM）的“Off-On”型荧光探针（ 图1 图1 ），对甲基硫菌灵的检出限为7.6 nmol/L，且其他干扰离子及农药的存在均不会影响探针的响应。 基于金属离子淬灭的CDs荧光探针具有成本低、效率高等优点，但其在实际应用中存在如下缺陷：1） 在传感平台构建之前，农药、金属离子及CDs之间的相互作用强度不可预测；2） 农药与金属离子的相互作用较弱，导致荧光恢复效率较低，可能使检测灵敏度下降；3） 金属离子的使用可能会产生一些副反应，从而影响传感平台的性能。 2.2　基于纳米颗粒的CDs荧光探针 2 纳米颗粒是指尺寸在1~100 nm之间的一种超小粒子，具有独特的光、电、磁及热的特性。除CDs外，半导体量子点、贵金属纳米颗粒（纳米金、纳米银）、贵金属纳米团簇均在农药检领域有深入的研究 ［ 19 19 ~ 22 22 19~22 ］ 。通过选择、组合、利用多种纳米颗粒的特征，可以设计杂化纳米材料，用于构建较高检测灵敏度、选择性及实用性的荧光探针。纳米颗粒通常可以作为CDs的荧光淬灭剂、增强剂及比率型传感的底物。 常规的纳米颗粒可以有效地淬灭CDs的荧光，也可以与多种不同的农药相互作用，使CDs的荧光恢复。Wang等 ［ 23 23 ］ 发现羊毛衍生CDs的荧光可以通过内过滤效应被银纳米颗粒（Ag NPs）淬灭，而草甘膦（glyphosate）可以诱导银纳米颗粒的聚集，使CDs的荧光恢复，据此构建了对残留草甘膦的定量检测方法（ 图2 图2 ）。在最佳实验条件下，草甘膦的线性检测范围为0.025~2.500 μg/mL，检出限为12 ng/mL。Sajwan等 ［ 24 24 ］ 同样基于金纳米颗粒（Au NPs）对CDs的内过滤效应，构建了一种快速、灵敏且简便的检测涕灭威的荧光传感平台。随着CDs的加入，金纳米颗粒在静电作用力的诱导下发生聚集，使CDs的荧光被淬灭；而随着涕灭威的加入，金纳米颗粒通过Au—N键或Au—S键与涕灭威发生相互作用，在CDs与金纳米颗粒之间形成一个插入层，减弱了金纳米颗粒的内过滤效应，使CDs荧光恢复。该方法的线性检测范围为3.8~76.0 μg/L，检出限为3.02 μg/L。 其他纳米颗粒也可以作为CDs传感系统中的信号增强剂，获得较高的荧光检测灵敏度。这种信号增强可能来源于金属纳米颗粒附近的局域场增强效应，即通过非辐射相互作用或辐射衰减工程介导的等离子体耦合效应，这类效应统称为金属增强荧光（metal-enhanced fluorescence，MEF）效应 ［ 25 25 ］ 。最近， Jiménez-López等 ［ 26 26 ］ 利用石墨烯量子点（graphene quantum dots，GQDs）以及半胱氨酸包裹的银纳米颗粒构建了一种用于草甘膦检测的分析平台。由于GQDs与银纳米颗粒表面的等离子耦合作用，导致银纳米颗粒辐射衰减速率以及荧光发射的增强，而草甘膦的加入能淬灭GQDs-银纳米颗粒复合系统的荧光，从而实现对草甘膦的灵敏检测，检出限低至9 ng/mL。然而MEF效应的机制非常复杂，且目前MEF效应的机理尚不清晰，限制了基于MEF及CDs的荧光探针的设计开发。荧光共振能量转移（FRET）效应也是一个非辐射的能量转移过程，主要通过供体/受体分子之间的偶极相互作用，将能量从供体荧光分子传递给邻近的受体分子，使供体荧光强度下降及受体荧光强度增加 ［ 12 12 ］ 。研究者 ［ 27 27 ］ 利用CdTe量子点（能量受体）及CDs（能量供体）构建了一种基于FRET效应的荧光探针（ 图3 图3 ），可以高灵敏、选择性地检测绿麦隆（chlortoluron）。在实验中，首先发生从CDs到CdTe量子点的荧光共振能量转移，使CdTe量子点荧光增强，同时CDs的荧光被淬灭；但由于绿麦隆与CdTe量子点之间的相互作用力更强，所以随着绿麦隆的加入，CdTe量子点的荧光被淬灭，而CDs的荧光随之增强。在最佳实验条件下，该探针对农药的线性检测范围为0.24~85.00 nmol/L，检出限为0.078 nmol/L，常见的阳离子及阴离子不会对该农药的检测造成干扰。 2.3　基于天然酶的CDs荧光探针 2 天然酶是一种具有独特催化功能的特异性蛋白，在体内无毒且环境友好，可以高效地催化各种生物化学反应 ［ 28 28 ， 29 29 ］ 。酶对其催化底物具有很强的特异性，因此基于酶的生物传感器在分析领域吸引了广泛的关注 ［ 30 30 ］ 。在用于残留农药检测的基于酶的CDs荧光探针中，有两种主要的检测机制：1） 抑制机制，即农药通过阻断酶的活性位点抑制酶的活性，从而诱发荧光信号响应；2） 催化机制，即以农药作为酶的底物，影响催化过程，进而影响荧光信号响应。其中，基于酶抑制机制的CDs荧光探针在农药的分析检测中起到了十分重要的作用。 胆碱酯酶（cholinesterase，ChE）包括乙酰胆碱酯酶（AChE）及丁酰胆碱酯酶（BChE），是有机磷类、氨基甲酸酯类农药的监测中使用最广泛的两种酶 ［ 31 31 ］ 。有机磷中含有不同比例的C、N、O、S包围的磷酸酯键，可以通过亲核反应进攻胆碱酯酶的丝氨酸残基 ［ 7 7 ］ 。磷酸化的胆碱酯酶无法催化底物的水解以及抑制硫代胆碱的形成，而硫代胆碱可以竞争性地与Ellman试剂、金属离子及贵金属纳米颗粒相互作用，并直接/间接地调节其与CDs的相互作用。在所有用于残留农药检测的基于CDs及胆碱酯酶的分析策略中，硫代胆碱调控的策略在农药检测中具有较高的地位。 由于金属离子（如Cu 2+ 、Au 3+ ）与巯基（—SH）之间具有较强的亲和力，因此金属离子可以与硫代胆碱形成较强的配合物，用于残留农药检测的CDs荧光探针的构建 ［ 32 32 ， 33 33 ］ 。Hou等 ［ 32 32 ］ 基于Cu 2+ 与CDs的体系构建了一种用于敌敌畏（dichlorvos）检测的荧光探针（ 图4 图4 ）。首先CDs的荧光可以被Cu 2+ 选择性地淬灭；然后，当AChE及硫代乙酰胆碱存在时，AChE催化硫代乙酰胆碱水解，形成硫代胆碱，通过硫代胆碱与Cu 2+ 之间更强的亲和力，使CDs的荧光恢复。当AChE的抑制剂（敌敌畏）存在时，AChE无法催化硫代乙酰胆碱的水解，使CDs的荧光保持淬灭状态，从而实现对敌敌畏的定量检测。 还原性的Ellman试剂（5，5-二硫代二苯（2-硝基苯甲酸），DTNB）也常用于残留农药检测的CDs荧光探针的构建。DTNB可以作为硫代胆碱的显色试剂，验证硫代乙酰胆碱的水解产物的形成，从而根据颜色信息的变化定量计算残留农药的浓度 ［ 34 34 ， 35 35 ］ 。根据这一特性，Li等 ［ 34 34 ］ 基于AChE对 CDs荧光淬灭的调控，构建了一种无标记的残留有机磷农药检测的双模式（荧光、比色）平台。AChE可以催化硫代乙酰胆碱水解形成硫代胆碱，进而使DTNB降解生成黄色的5-硫代-2-硝基苯甲酸（TNBA）。而带正电的TNBA可以吸附在CDs表面，通过动态淬灭的方式淬灭CDs的荧光。当有机磷农药存在时，AchE的活性被抑制，从而使CDs的荧光仍然保持。在最佳实验条件下，该检测平台对对氧磷的检出限为0.4 ng/mL。然而， AChE对有机磷农药的选择性较差，使该检测平台在检测实际样品时受到了一定的限制。 将CDs与其他功能性纳米材料结合也是一种构建理想探针的有效策略 ［ 36 36 ］ 。Chen等 ［ 37 37 ］ 通过水热法制备了一种荧光发射波长位于490 nm的CDs探针，其荧光强度会被金纳米颗粒有效地淬灭。而AChE催化硫代乙酰胆碱生成的硫代胆碱，会诱导金纳米颗粒的聚集，伴随着其在520 nm处吸收峰的下降，并最终使CDs的荧光恢复。而农药西维因的存在会抑制AChE的活性以及金纳米颗粒的聚集，使CDs荧光保持淬灭的状态。在最佳实验条件下，该方法的线性检测范围为0.20~150.00 μg/mL。Yan等 ［ 38 38 ］ 基于CDs和MnO 2 纳米片成功实现了对有机磷农药的定量检测。首先对3-氨基苯硼酸进行水热处理得到荧光CDs，再用MnO 2 纳米片通过FRET效应淬灭CDs的荧光。在BChE及硫代乙酰胆碱存在时，酶的水解产物（硫代胆碱）可以高效诱发MnO 2 纳米片的降解，从而使CDs荧光恢复。而有机磷作为酶的抑制剂，可以阻止硫代胆碱的形成及MnO 2 纳米片的降解，使CDs的荧光保持淬灭的状态。在最佳实验条件下，该方法可以有效地检测对氧磷，线性检测范围为0.05~5.00 ng/mL，检出限为0.015 ng/mL。Huang等 ［ 39 39 ］ 构建了一种用于残留农药检测的Cu 2+ 及CDs/CdTe量子点杂化水凝胶的比率型探针（ 图5 图5 ）。其原理为：CdTe量子点的荧光可以被Cu 2+ 动态淬灭，而CDs的荧光对Cu 2+ 无响应，同时AChE催化乙酰硫代胆碱水解形成的硫代胆碱（TCh），可以通过强的Cu—S键与Cu 2+ 结合，从而使CdTe量子点的荧光恢复。当酶的竞争性抑制剂（敌敌畏）存在时，会抑制硫代胆碱-Cu复合物的形成，从而改变CDs的比率型荧光信号。这种可携带的诊断试剂盒的线性检测范围为1~40 ng/mL，检出限为0.38 ng/mL。 除使用单一酶外，也可以采取多种酶联用的方法构建残留农药检测的CDs荧光探针。由于胆碱氧化酶（ChOx）的高催化活性及高选择性，基于ChOx的多种酶体系在残留农药检测的CDs荧光探针构建方面也有广泛应用 ［ 40 40 ］ 。通常情况下，胆碱酯酶可以催化乙酰胆碱水解产生胆碱，作为ChOx的底物催化产生H 2 O 2 及甜菜碱 ［ 41 41 ］ 。通过检测H 2 O 2 的荧光探针可以监测整个生物催化过程。Sahub等 ［ 42 42 ］ 构建了一种用于监测有机磷农药的GQDs/酶的纳米检测平台（ 图6 图6 ）。AChE及ChOx的酶联反应会产生一定量的H 2 O 2 ，H 2 O 2 可以有效淬灭GQDs的荧光，而有机磷农药又会使GQDs的荧光恢复，且荧光的恢复与有机磷农药的浓度相关。在最佳实验条件下，GQDs/AChE/ChOx对有机磷的检出限为0.172 μg/mL。由于单独的酶对相应的底物具有较好的特异性，所以多种酶联用的检测平台具有较好的抗干扰能力。但该检测策略存在反应时间较长、反应条件也更加苛刻的问题。 除了抑制策略之外，利用有机磷水解酶（organophosphorus hydrolase，OPH）对农药的催化作用也可以检测残留农药。有机磷农药可作为OPH的底物，因此通过定量分析OPH催化转化的产物，对有机磷农药进行定量分析。OPH是一种酰胺水解酶家族的脯氨酰氨基酸酶 ［ 43 43 ］ ，可以催化水解多种农药（如二嗪农及甲基对硫磷）。由于水解反应的不可逆性，故该反应的催化转化效率较高，但同时酶催化水解产物的活性也较高，很容易引发荧光信号的改变。因此，将OPH与CDs结合在一起，可用于检测甲基对硫磷。甲基对硫磷的水解产物（ p -硝基苯酚）能淬灭CDs的荧光，可据此建立残留甲基对硫磷的检测方法 ［ 44 44 ］ 。OPH对目标农药的特异性识别和降解功能对于发展绿色探针非常重要。然而，与AChE比较，OPH的催化机制尚不明确且其催化活性较差，检测的灵敏度也低于基于抑制机制的CDs荧光探针。 2.4　基于抗体的CDs荧光探针 2 抗体免疫球蛋白（immunoglobulin G，IgG）是一种由外源性物质（抗原）的刺激产生的具有保护性功能的蛋白，基于抗体的竞争性结合反应，广泛地应用于各种疾病的预防、诊断及治疗 ［ 45 45 ］ 。抗原与抗体之间的特异性识别可以作为生物识别元件，用于生物传感器件的构建 ［ 46 46 ， 47 47 ］ 。通常情况，需要CDs对抗体-抗原的免疫反应足够敏感，将免疫应答转变为荧光信号的淬灭或增强，通过“三明治”结构或直接/间接的竞争性原则构建可靠的免疫荧光探针。对农药分子进行衍生化修饰，可以使其满足免疫荧光检测的需求，但步骤非常繁琐且低效。因此，直接或间接的竞争性策略在设计用于残留农药检测的CDs及抗体的免疫荧光探针中非常有效 ［ 48 48 ］ 。Wang等 ［ 49 49 ］ 基于免疫荧光策略构建了一种针对草甘膦（glyphosate， GLY）的荧光检测方法（ 图7 图7 ）。他们将环境友好的CDs与草甘膦的抗体偶联在一起形成IgG-CDs，可以特异性地识别草甘膦。而IgG-CDs可以原位地对植物组织中草甘膦的分布进行分析。考虑到多余的IgG-CDs可能对草甘膦的检测产生影响，他们进一步构建了一种基于磁性Fe 3 O 4 及草甘膦的抗原磁性微球用于偶联多余的IgG-CDs，实现了对草甘膦的检测，线性检测范围为0.01~80.00 μg/mL，检出限为8 ng/mL，在实际样品（如河水、茶叶及土壤）中的回收率为87.4%~103.7%。 用于残留农药检测的CDs及抗体免疫荧光探针具有非常多的优越性，但也存在如下缺陷：1） 抗体与CDs的偶联效率较低；2） 免疫荧光检测的耗时较长；3） 抗体的稳定性有待进一步提高。这些缺陷限制了它在快速分析领域的应用。 3　 总结及展望 1 全球范围内的农药污染问题日益严重，促进了残留农药检测技术的迅速发展。虽然已有文献总结了用于残留农药检测的分析技术，但基于CDs荧光的检测方法仍有许多发展空间。CDs具有非常优异的荧光性质，突出的生物相容性以及良好的水溶性，是开发高灵敏残留农药检测技术的理想材料。CDs作为发光团、识别原件或催化剂，可以单独或与其他材料联合用于构建检测残留农药的精确探针。通过调节合成方法、条件，可以调控CDs的性质，开发多种多样的荧光探针。 基于已有的研究，用于残留农药检测的CDs荧光探针的研究主要面临如下挑战： 1） 高发光性能CDs的开发应用仍然较为缺乏。虽然目前许多工作报道了CDs具有近红外发光、磷光以及上转换荧光的特性，但这些特性很少用于相关探针的构建。近红外荧光、磷光及上转换荧光CDs的使用可以有效消除复杂生物基质中的背景干扰，极大地提高其在实际样品分析中的准确性。另外，将CDs与其他荧光纳米材料联合使用，构建双发射的比率型荧光探针，可以提供内置校准功能，减少仪器及环境因素的系统误差。 2） CDs的发光机制仍然存在一定的争议。关于CDs发光机制的研究有限，在一定程度上限制了CDs在残留农药检测领域的广泛应用。因此，全面、深入地研究CDs的发光机制对指导CDs的设计合成，提高其检测残留农药的性能具有重要意义。 3） 严重依赖大型荧光检测设备。目前，用于残留农药检测的CDs荧光探针严重依赖大型的荧光检测设备，对农药的实时、原位检测造成障碍。因此，需要开发便携性的微型化荧光检测设备，以方便用于不同领域实际样品中的残留农药检测。 总之，在未来的研究中，研究者们需要从不同的传感机制和合成策略中提取有益的思路，努力开发现有CDs的潜力及优势，实现残留农药的实时、快速、原位、高灵敏检测。 参考文献 FANG L ，
